Abstract Summary

Podoconiosis, a neglected tropical disease caused by prolonged exposure to red clay soil, leads to chronic leg swelling in poor tropical communities. Despite its significant impact, the disease’s mechanisms remain unclear, hampering diagnosis and treatment. This review examines recent progress in understanding genetic, immune, and tissue factors behind podoconiosis, while highlighting challenges in misdiagnosis and the urgent need for biomarkers and standardized tests.
